Dear Melissa:
Luminescence problems in LSC samples should be an indication of chemical
reactions in your cocktail-sample mixture. So you need to consider
everything that goes into the vial including some kind of contamination
inside the "new" and unused vials. Oxidizing agents and enzymes are
particularly good at producing luminescence.
I know you probably already know all this, but I hope you find the problem
and let us know what it was.
